目的:探索骨膜蛋白(periostin,POSTN)在乳腺癌组织、癌旁组织、乳腺正常组织中的表达情况及其临床意义。方法:选取昆明医科大学第三附属医院乳腺科2012年5月至2016年12月手术切除的45例乳腺癌患者的癌组织、癌旁组织及正常乳腺组织,另取15例患者手术切除的无菌癌组织及正常组织进行原代成纤维细胞培养,H-E及免疫组化染色鉴定原代培养的成纤维细胞。采用实时定量PCR(qPCR)分别分析乳腺癌组织、癌旁组织和乳腺正常组织以及癌相关成纤维细胞和正常成纤维细胞中POSTN mRNA的表达情况,并用相关性分析乳腺癌组织中POSTN表达水平和乳腺癌患者临床病理特征的关系。乳腺癌组织芯片H-E及免疫组化染色检测POSTN蛋白在乳腺癌组织中的表达定位。结果:乳腺癌组织POSTN mRNA表达水平显著高于癌旁组织和正常乳腺组织(P<0.05);癌相关成纤维细胞POSTN mRNA表达与正常成纤维细胞无明显差异(P>0.05)。POSTN蛋白主要在乳腺癌实质组织中表达,间质中表达较少。乳腺癌组织中POSTN mRNA表达水平与乳腺癌患者雌激素受体呈负相关(P<0.05)。结论:POSTN主要在乳腺癌组织实质中表达,其在乳腺癌组织中高表达可能与乳腺癌的发生发展相关。
Objective: To investigate the expression and clinical significance of periostin (POSTN) in breast cancer tissues, paracancerous tissues and normal breast tissues. Methods: Forty-five patients with breast cancer who underwent surgical resection from May 2012 to December 2016 in Department of Breast Surgery, the Third Affiliated Hospital of Kunming Medical University were enrolled in this study. Fifteen patients underwent surgical excision The primary fibroblasts were cultured in bacterial and normal tissues. Primary cultured fibroblasts were identified by HE and immunohistochemical staining. The expression of POSTN mRNA in breast cancer tissues, paracancerous tissues and normal breast tissues as well as in cancer-associated fibroblasts and normal fibroblasts were analyzed by real-time quantitative PCR (qPCR). The correlation between the expression of POSTN in breast cancer tissues and Relationship between clinicopathological characteristics of breast cancer patients. Detection of breast cancer tissue H-E and immunohistochemical staining for the expression of POSTN protein in breast cancer. Results: The expression of POSTN mRNA in breast cancer tissues was significantly higher than that in paracancerous tissues and normal breast tissues (P <0.05). There was no significant difference in the expression of POSTN mRNA between normal fibroblasts and normal fibroblasts (P> 0.05). POSTN protein is mainly expressed in breast parenchyma and less in interstitial tissue. The expression of POSTN mRNA in breast cancer tissues was negatively correlated with estrogen receptor in breast cancer (P <0.05). Conclusion: The expression of POSTN mainly in breast cancer tissue and its high expression in breast cancer may be related to the occurrence and development of breast cancer.
