目的:观察淋球菌孔蛋白疫苗免疫日本大耳白兔后所诱导的免疫应答。方法:兔背皮下注射经原核构建表达并纯化的淋球菌孔蛋白B,ELISA动态检测免疫后大耳兔血清抗体滴度的变化及最高效价;后腿皮内注射孔蛋白B,观察迟发型超敏反应(DTH)、免疫血清体外玻片凝集及抗菌作用。结果:初次免疫后2周,大耳兔体内已产生抗体,但效价较低;第2次免疫后2周血清效价达到1∶2000;第3次免疫后2周血清抗体水平成倍升高,效价为1∶4000;第4次免疫后2周血清最高效价达到1∶12000。大耳兔腿部发生DTH反应,出现明显红肿,于注射后第4天消退。血清体外凝集试验呈阳性,加入补体后发生淋球菌菌体裂解。结论:淋球菌孔蛋白疫苗可诱发动物特异性免疫应答,这为淋病疫苗的进一步研究提供了理论和实验依据。
Objective: To observe the immune response induced by gonococcal porcine immunization of Japanese white rabbits. Methods: The gonococcal porin B expressed and purified by pronucleus was subcutaneously injected into rabbits. ELISA was used to detect the change of antibody titers and the highest titer of the rabbit serum after immunization. The posterior leg intradermal injection of porin B was used to observe the delayed type Hypersensitivity reaction (DTH), immune serum in vitro slide agglutination and antibacterial effects. Results: Two weeks after the first immunization, antibodies were produced in the large-eared rabbits, but the titers were low. The serum titer reached 1: 2000 at 2 weeks after the second immunization. Serum antibody levels doubled at 2 weeks after the third immunization High, the titer is 1: 4000; the highest titer of the serum reaches 1: 12000 after 2 weeks of the 4th immunization. The DTH reaction occurred in the legs of the large-eared rabbit, with obvious redness and swelling, which subsided on the 4th day after injection. Serum in vitro agglutination test was positive, adding Neisseria gonorrhoeae bacterial lysate after cell lysis. Conclusion: Neisseria gonorrhoeae porin vaccine can induce animal specific immune response, which provides theoretical and experimental evidence for further study of gonorrhea vaccine.
