目的探讨外周血干细胞亚群CD+34/CD-38与CFU蛳GM,BFU蛳E的相关性及其影响因素。方法应用流式细胞仪测定外周血干细胞冻存前后的CD+34及CD+34/CD-38,对冻存前后的PBSC进行CFU蛳GM,BFU蛳E测定。对数据进行对比分析及相关性分析。结果冻存后较冻存前集落产率明显降低(P<0.05),而冻存1个月与3个月的集落产率差异无显著性(P>0.05)。冻存前后CD+34/CD-38百分率与CFU蛳GM、BFU蛳E的数目相关性分析显示二者均存在显著相关性。结论外周血干细胞CD+34/CD-38百分率与CFU蛳GM,BFU蛳E集落产率之间存在显著相关性,二者可作为测定PBSC质与量的互补指标。冷冻保存时间对PBSC的膜分子抗原无明显影响。冷冻保存减少了CFU蛳GM,BFU蛳E的产率,但冻存1个月与3个月对PBSC的集落产率影响不明显。
Objective To investigate the correlation between peripheral blood stem cell subsets CD+34/CD-38 and CFU蛳GM, BFU蛳E and its influencing factors. Methods Flow cytometry was used to measure CD34+ and CD+34/CD-38 before and after cryopreservation of peripheral blood stem cells. CFU蛳GM and BFU蛳E were measured in PBSC before and after cryopreservation. Compare the data and analyze the correlation. Results Compared with cryopreservation, the yield of colonies before cryopreservation was significantly lower (P<0.05), but there was no significant difference in the colony yield between cryopreserved one and three months (P>0.05). The correlation between the percentage of CD+34/CD-38 and the numbers of CFU蛳GM and BFU蛳E before and after cryopreservation showed that there was a significant correlation between the two. Conclusion There is a significant correlation between the percentage of CD34/CD-38 in peripheral blood stem cells and the CFU蛳GM and BFU蛳E colony yields, both of which can be used as complementary indicators to determine the quality and quantity of PBSC. The cryopreservation time had no significant effect on the membrane antigens of PBSC. Cryopreservation reduced the yields of CFU蛳GM and BFU蛳E, but cryopreservation for 1 month and 3 months had no significant effect on the PBSC colony yield.
