利用短期诱导实验 ,分析了花生四烯酸对红豆杉细胞培养中紫杉烷合成的影响。在适宜浓度的花生四烯酸诱导下 ,紫杉醇 (taxol)的产量提高了近 3倍 ,同时 10 -去乙酰基巴卡亭 (10 - DAB)与巴卡亭 (baccatin )相应上升。通过对紫杉醇合成代谢途径的等拓扑结构简化与动力学分析 ,获得了诱导子加入后可能的 3种中间代谢产物浓度转移模式。结合花生四烯酸短期诱导实验结果 ,提出了花生四烯酸的加入提高了 10 - DAB合成通量的观点 ,诱导子作用位点的确定为多诱导子配伍方案奠定了基础
Using short-term induction experiments, the effect of arachidonic acid on the synthesis of taxane in cell culture of Taxus chinensis was analyzed. At the appropriate concentration of arachidonic acid induction, taxol production increased nearly 3-fold, while 10-deacetylbaccatin (10-DAB) increased in response to baccatin. Through isotopological structural simplification and kinetic analysis of paclitaxel anabolic pathways, the possible intermediate metabolite concentration transfer patterns after the elicitor was added were obtained. Combined with the results of short-term induction experiments of arachidonic acid, it was proposed that the addition of arachidonic acid improves the synthetic flux of 10-DAB, and the determination of the site of elicitors establishes the basis for the compatibility of multiple inducers.
