目的:利用响应面分析法对黄连生物碱的提取工艺进行优化。方法:以黄连生物碱提取率为指标,采用Box-Behnken中心组和设计建立数学模型,通过对超声时间、乙醇浓度、回流提取时间及液料比4个影响因素的考察,得出各因素对黄连生物碱提取率的影响程度。结果:优化后黄连生物碱的最佳提取工艺为:加70%乙醇,液料比16.5∶1(m L/g),超声时间27 min,回流提取时间1.5 h,黄连生物碱理论提取率为255.475 mg/g,实际测得提取率为252.897 mg/g。结论:Box-Behnken实验设计方法可以较好的对黄连总生物碱提取工艺进行优化。
Objective: To optimize the extraction process of berberine alkaloids by response surface methodology. Methods: Taking the extraction rate of alkaloids from Coptis chinensis as an index and using Box-Behnken central group and the design of mathematical model, the factors such as ultrasound time, ethanol concentration, reflux extraction time and liquid to solid ratio were investigated. Coptis alkaloid extraction rate of the degree of influence. Results: The optimal extraction process of the alkaloids from Coptis chinensis Franch was 70% ethanol, the ratio of liquid to material was 16.5:1 (m L / g), the ultrasonic time was 27 min and the time of reflux extraction was 1.5 h. The theoretical extraction rate of Coptis alkaloids was 255.475 mg / g, the actual measured extraction rate was 252.897 mg / g. Conclusion: The Box-Behnken experimental design method can optimize the extraction process of total alkaloids from Coptis chinensis.
