目的探讨补肾富钙中药对大鼠卵巢辐射损伤所致骨质疏松形态计量学的影响。方法40只SD大鼠随机分为四组(n=10),分别为假手术组(灌服1%纤维素90 d)、模型组(以50 Gyγ射线局部照射大鼠卵巢建立卵巢辐射损伤模型,1%纤维素灌服90 d)、尼尔雌醇组(造模后给予尼尔雌醇灌胃90 d)和补肾富钙中药组(造模后给予补肾富钙中药灌胃90 d)。各组大鼠于处死前第14天和第4天分别腹腔注射盐酸四环素(30 mg/kg)进行标记,处死后取右侧股骨组织和L4腰椎组织分别进行骨组织形态计量和扫描电镜观察。结果与模型组比较,补肾富钙中药组和尼尔雌醇组的骨小梁体积显著增加,骨吸收表面减少;尼尔雌醇组的骨形成表面和矿化沉积率下降,但在补肾富钙中药组则无显著变化。扫描电镜观察显示,模型组骨小梁变细、中断、排列稀疏,而补肾富钙中药组和尼尔雌醇组上述变化较轻微。结论补肾富钙中药可有效预防卵巢辐射损伤诱发的骨质疏松。
Objective To investigate the effect of kidney-enriching and calcium-rich traditional Chinese medicine on the morphometrics of osteoporosis induced by irradiation damage to the ovary in rats. Methods Forty Sprague-Dawley rats were randomly divided into four groups (n=10): sham operation group (perfused with 1% cellulose for 90 days) and model group (Ovarian radiation damage model was established by irradiation of rat ovary with 50 Gy γ-rays). ,1% cellulose fed for 90 d), Nilestriol group (given Nilestriol 90 days after modeling) and kidney-enriched calcium group (after kidney-enriched-calcium-enriched Chinese herbs were given for 90 d after modeling) . Rats in each group were given intraperitoneal injection of tetracycline hydrochloride (30 mg/kg) on the 14th and the 4th day before being sacrificed. After the death, the right femur tissue and L4 lumbar vertebrae were taken for bone histomorphometry and scanning electron microscopy. Results Compared with the model group, the trabecular bone volume of the Bushen-Cai-he-rich Chinese medicine group and the Nylestriol group significantly increased, and the bone resorption surface decreased; the bone formation surface and mineralization deposition rate of the Nystriol group decreased, but it was in the kidney-rich There was no significant change in the calcium Chinese medicine group. Scanning electron microscopy showed that the trabecular bone in the model group was thin, interrupted, and sparsely arranged, while the above changes in the kidney-enriched-calcium-enriched Chinese medicine group and the nilestriol group were slight. Conclusion The kidney-enriched and calcium-rich traditional Chinese medicine can effectively prevent osteoporosis induced by ovarian radiation damage.
