将二吡啶基乙基胺(DPA)与芘缩合还原后得到1-(芘-1-基)-N,N-双(吡啶-2-基甲基)乙胺(PYPA)可用作Zn~(2+)探针.在焦磷酸(PPi)存在下Me OH/H2O(3∶2,V/V,p H 7.20,HEPES 1.0 mmol/L)缓冲液中PYPA对Zn~(2+)显示出高度专一性,加Zn~(2+)后20 s在383 nm处荧光发射可达最大值,且检测不受各种阳离子或阴离子的干扰,其检测极限可低至30 nmol/L.活细胞成像研究表明PYPA可用于人肝细胞L-02中Zn~(2+)荧光标记.
(Pyrene-1-yl) -N, N-bis (pyridin-2-ylmethyl) ethylamine (PYPA) is obtained after the condensation of dipyridylethylamine (DPA) (2+) PYPA in the presence of pyrophosphate (PPi) in Me OH / H 2 O (3: 2, V / V, p H 7.20, HEPES 1.0 mmol / L) Highly specific, fluorescence emission reached its maximum at 383 nm in 20 s after addition of Zn 2+, and its detection was not disturbed by various cations or anions. Its detection limit was as low as 30 nmol / L. Live cell imaging studies indicate that PYPA can be used to label Zn 2+ in human hepatocytes L-02.
