目的:建立天然牛黄HPLC-ELSD特征图谱并与酶促牛黄特征图谱进行比较。方法:采用HPLC-ELSD法,色谱条件:Diamonsil C18柱(250 mm×4.6 mm,5μm);以甲醇和0.1%的醋酸水溶液为流动相,流速0.8 mL.min-1,梯度洗脱,漂移管温度110℃,气体流速2.0 L.min-1;分析时间60 min。结果:在选定的色谱条件下建立了天然牛黄色谱特征图谱共有模式,并对酶促牛黄与天然牛黄的特征图谱进行比较。结论:所建立的天然牛黄HPLC-ELSD特征图谱方法精密度高、重现性好,可用于酶促牛黄的鉴别研究。
OBJECTIVE: To establish a HPLC-ELSD characterization map of natural bezoar and compare it with that of enzymatic bezoar. Methods: The HPLC-ELSD method was used. The chromatographic conditions were Diamonsil C18 column (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of methanol and 0.1% aqueous acetic acid with a flow rate of 0.8 mL · min- Temperature 110 ℃, gas flow 2.0 L.min-1; analysis time 60 min. Results: The common pattern of natural bezoar color spectrum was established under the selected chromatographic conditions, and the characteristic patterns of enzymatic bezoar and natural bezoar were compared. Conclusion: The established HPLC-ELSD characteristic curve method of natural bezoar has high precision and reproducibility and can be used for the identification of enzymatic bezoar.
