目的研究粉防己碱对糖尿病(DM)大鼠心肌的保护作用及相关机制。方法36只W istar大鼠用链脲佐菌素(STZ)诱发糖尿病模型后,随机分为粉防己碱治疗组、苯那普利治疗组及糖尿病未治疗组,以10只正常大鼠作为对照。12 w末检测血糖、血脂、心肌组织血管紧张素Ⅱ,应用RT-PCR检测心肌结缔组织生长因子(connective tissue growth factor,CTGR)mRNA的表达水平。结果DM未治疗组心肌组织血管紧张素Ⅱ含量、CTGR mRNA的表达分别为(26.5±3.38)和(1.0±0.10),明显高于对照组和治疗组(P<0.01)。结论粉防己碱能在一定程度上降低心肌血管紧张素Ⅱ含量,抑制CTGF mRNA表达,从而可能减轻糖尿病性心肌病的病理变化。
Objective To study the protective effect of tetrandrine on the myocardium of diabetic rats and its related mechanism. Methods Thirty-six Wistar rats were induced with streptozotocin (STZ) and randomly divided into tetrandrine treatment group, benazepril treatment group and untreated diabetic group. Ten normal rats were used as controls. . At the end of the 12th day, blood glucose, blood lipids, and myocardial tissue angiotensin II were detected. RT-PCR was used to detect the expression level of cardiac connective tissue growth factor (CTGR) mRNA. Results The levels of angiotensin II and CTGR mRNA in the untreated group were (26.5±3.38) and (1.0±0.10), which were significantly higher than those in the control and treatment groups (P<0.01). Conclusion Tetrandrine can reduce the content of angiotensin II in myocardium to a certain extent, inhibit the expression of CTGF mRNA, which may reduce the pathological changes of diabetic cardiomyopathy.
