失重或模拟失重引起体液的头向转移,导致上半身血容量增加,从而引起心房肌心房利钠肽(atrial natriuretic peptide,ANP)分泌的改变,但现有研究报道缺乏一致的结论。本研究拟观测模拟失重大鼠心肌ANP表达的变化,并以可溶性N-乙基马来酰亚胺敏感因子附着蛋白受体(soluble N-ethylmaleimide-sensitive factor attachment protein receptors,SNARE)家族相关蛋白的变化作为ANP变化的佐证。采用公认的尾部悬吊大鼠模型在地面模拟失重状态,蛋白印迹法检测心房与心室肌ANP与SNARE相关蛋白的表达水平,并进行半定量分析。结果显示,与同步对照组(CON)相比,尾部悬吊4周大鼠(SUS)心房肌ANP表达增加;在CON大鼠左心室肌仅显示微量ANP表达,但SUS大鼠心室肌ANP表达明显增加。相比CON组,SUS组囊泡SNARE(v-SNARE)VAMP-1/2在心房肌与心室肌表达明显增加;细胞膜靶标SNARE(t-SNARE)syntaxin-4的表达未改变,但SNAP-23在心房肌的表达明显增加。SNARE调节蛋白synip与Munc-18c的表达在心房肌与心室肌未发生显著性改变。上述结果表明,尾部悬吊4周引起心房肌ANP表达增加,心室肌呈现ANP表达;与ANP囊泡相关的VAMP-1/2的表达亦增加,佐证心房肌与心室肌ANP表达增加。
Loss of body mass or simulated weightlessness cause head-to-body fluid transfer, leading to an increase in blood volume in the upper body resulting in changes in atrial natriuretic peptide (ANP) secretion in the atrium, but no consistent conclusions have been reported in previous studies. This study was designed to observe the changes of ANP expression in myocardium of simulated weightless rats and to investigate the effects of soluble N-ethylmaleimide-sensitive factor attachment protein receptors (SNARE) family of related proteins Changes as evidence of ANP changes. The rat model of tail suspension was used to simulate weightlessness on the ground. The protein expression of ANP and SNARE in atrial and ventricular myocardium was detected by Western blot and semi-quantitative analysis was performed. The results showed that the expression of ANP increased in the atrial myocardium of the tail suspension for 4 weeks (SUS) compared with that in the synchronous control group (CON); only ANP was expressed in the left ventricular muscle of CON rats, but ANP in the SUS rat ventricular myocytes obviously increase.
