目的:观察祛浊通痹方对高尿酸模型大鼠血尿酸及黄嘌呤氧化酶(xanthine oxidase,XOD)的影响,阐释祛浊通痹方治疗痛风改善尿酸代谢的作用机制。方法:通过喂养高钙饲料和腺嘌呤混悬液,制作大鼠痛风高尿酸血症模型,然后分为别嘌醇组(造模后灌胃别嘌醇)、中药组(造模后灌胃祛浊通痹方)、模型组(造模后不用药),另取正常大鼠为空白组(不造模不用药),分别于用药前及14d后取血检测各组血尿酸、黄嘌呤氧化酶水平。结果:痛风高尿酸血症模型大鼠血尿酸水平显著高于空白组(P<0.01);与治疗前比较,别嘌醇组、中药组血尿酸及XOD均明显下降(P<0.01)。治疗后各组间比较,模型组血尿酸及XOD高于别嘌醇组及中药组(P<0.01);别嘌醇组的XOD低于中药组(P<0.01)。结论:喂养高钙饲料和腺嘌呤混悬液能成功制作大鼠痛风高尿酸血症模型,祛浊通痹方能显著降低尿酸和抑制XOD的活性。
Objective: To observe the effect of Quzhuotongbi prescription on blood uric acid and xanthine oxidase (XOD) in rats with hyperuricemia and to explain the mechanism of Quxutongbi treatment of gout to improve uric acid metabolism. Methods: Gout hyperuricemia model was made by feeding high-calcium diet and adenine suspension, then divided into allopurinol group (intragastric administration of allopurinol after modeling), Chinese medicine group The rats in the model group (no drug after modeling) and the normal rats as the blank group (no model without drug) were taken before and 14 days after treatment respectively to detect blood uric acid, xanthine Oxidase levels. Results: The level of serum uric acid in the rats with gout hyperuricemia was significantly higher than that in the blank group (P <0.01). Compared with before treatment, the levels of serum uric acid and XOD in allopurinol and Chinese herbs groups were significantly decreased (P <0.01). After treatment, the levels of serum uric acid and XOD in the model group were higher than those in the allopurinol group and the traditional Chinese medicine group (P <0.01). The XOD in the allopurinol group was lower than that in the traditional Chinese medicine group (P <0.01). Conclusion: The model of hyperuricemia induced by gout in rats can be successfully produced by feeding high-calcium diet and adenine suspension, and removing uric acid and inhibiting the activity of XOD can be significantly reduced by Quzuo Tongbi.
