利用SRAP标记和SSR标记对柳枝稷杂交种进行鉴定,比较2种标记在柳枝稷杂种鉴定方面的效率和准确性,为柳枝稷乃至异花授粉多倍体植物的杂种真实性鉴定提供科学依据。分别利用SRAP与SSR标记技术对柳枝稷165个杂交种单株和亲本DNA进行扩增,分析杂交种与亲本扩增的多态性条带数,并鉴别真假杂交种。与SRAP标记相比,共显性的SSR标记具有高效性,本研究中仅用1对SSR引物就鉴别了所有杂交后代的真实性,多态性条带比率为100%。而显性SRAP标记用了6对才能鉴别所有杂交后代的真实性,多态性条带比率为40%,相对较低。
The SRAP and SSR markers were used to identify the switchgrass hybrids. The efficiency and accuracy of the two markers in the identification of the switchgrass hybrids were compared to provide a scientific basis for the authenticity identification of hybrids of the switchgrass and even cross pollination polyploid plants. SRAP and SSR markers were used to amplify the 165 plant species and parental DNAs of switchgrass. The number of polymorphic bands amplified by hybrids and their parents was analyzed, and the true and false hybrids were identified. Compared with the SRAP marker, the codominant SSR marker is highly efficient. In this study, only one pair of SSR primers was used to identify the authenticity of all the hybrids. The percentage of polymorphic bands was 100%. However, six pairs of dominant SRAP markers were used to identify the authenticity of all the progeny of hybrids. The percentage of polymorphic bands was 40%, which was relatively low.
