【目的】探究淫羊藿水提取物联合抗菌药对耐药大肠杆菌的体外抑菌效果,为兽医临床中西联合用药提供参考依据,也为中药在抑制耐药菌株上的开发研究打下基础。【方法】采用二倍微量肉汤稀释法测定淫羊藿水提取物及各抗菌药对产超广谱β-内酰胺酶(ESBLs)大肠杆菌的最小抑菌浓度(MIC),然后以微量肉汤棋盘稀释法测定淫羊藿水提取物与抗菌药联用后的部分抑菌浓度指数(FICI)。【结果】淫羊藿水提取物与粘杆菌素、头孢曲松、头孢他啶、头孢噻呋、阿莫西林、磷霉素、阿米卡星、氟苯尼考、乙酰甲喹、林可霉素联用后能有效降低药物的MIC,对应的MIC均降低4倍以上,联合用药的FICI分别为0.13、0.26、0.26、0.25、0.51、0.51、0.50、0.75、0.75和0.75,对产ESBLs大肠杆菌的抑制效果表现出协同或相加作用;以其联合诱导产ESBLs大肠杆菌传代,从第1代即能明显降低这些抗菌药的MIC,且稳定至第3代。【结论】淫羊藿水提取物与粘杆菌素、头孢他啶、头孢曲松、头孢噻呋、阿莫西林、磷霉素、阿米卡星、氟苯尼考、乙酰甲喹、林可霉素联用是通过协同或相加作用而提高抗菌药对产ESBLs大肠杆菌的抑制效果,但不宜与磺胺类、喹诺酮类联合用药。
【Objective】 To explore the antibacterial activity of Epimedium water extract combined with antibacterial agents on drug-resistant Escherichia coli in vitro and to provide a reference for veterinary clinical Chinese and Western medicine and lay a foundation for the development of traditional Chinese medicine in inhibiting drug-resistant strains. 【Method】 The minimum inhibitory concentration (MIC) of Epimedium pubescens water extract and each antibacterial agent against Escherichia coli producing extended-spectrum β-lactamases (ESBLs) was determined by double broth microdilution method. Then, Determination of partial antibacterial concentration index (FICI) after combination of Epimedium water extract with antibacterials by the chessboard dilution method. [Results] Epimedium water extract and colistin, ceftriaxone, ceftazidime, ceftiofur, amoxicillin, fosfomycin, amikacin, florfenicol, mequindox, lincomycin After combination, the MICs of the drugs can be effectively reduced, and the corresponding MICs are reduced by more than 4 times. The FICIs of the combination drugs are 0.13, 0.26, 0.26, 0.25, 0.51, 0.51, 0.50, 0.75, 0.75 and 0.75 respectively. The inhibitory effect showed a synergistic or additive effect. With the combination of induction of ESBLs-producing Escherichia coli passage, the MICs of these antibacterials were significantly reduced from the first generation and were stable to the third generation. 【Conclusion】 Epimedium water extract was effective against colistin, ceftazidime, ceftriaxone, ceftiofur, amoxicillin, fosfomycin, amikacin, florfenicol, mequindox, The synergistic effect is to enhance the inhibitory effect of antibacterials on ESBLs-producing Escherichia coli through synergistic or additive effects, but not combined with sulfonamides and quinolones.
