目的:从岩黄连的粗提物中快速分离生物碱类成分。方法:采用高速逆流色谱技术,以正丁醇-乙酸乙酯-水-甲酸(5∶1∶5∶0.01)和正丁醇-乙酸乙酯-甲醇-水-甲酸(5∶5∶1∶9∶0.05)作为两相溶剂系统分离岩黄连中的主要生物碱。结果:经过连续三次色谱分离,从岩黄连的正丁醇粗提物(300 mg)中得到6个纯度较高的生物碱,分别为:scoulerine(3.6 mg,质量分数:71%)、isocorydine(9.2 mg,质量分数:92%)、dehydrocheilanthifoline(5.5 mg,质量分数:85%)、dehydrocavidine(7.5 mg,质量分数:76%)、palmatine(20.4 mg,质量分数:90%)、berberine(20.9 mg,质量分数:97%)。结论:采用高速逆流色谱法制备分离岩黄连中的生物碱,省时、方便、产物得率和纯度较高。
OBJECTIVE: To rapidly separate alkaloids from the crude extracts of Rhizoma Coptidis. Methods: High-speed countercurrent chromatography was used for the determination of n-butanol, ethyl acetate and water-formic acid (5:1:5:0.01) and n-butanol-ethyl acetate- : 0.05) as a two-phase solvent system to separate the main alkaloids in the genus Rhizoma Coptidis. Results: After three consecutive chromatographic separations, six highly pure alkaloids were obtained from crude n - Butanol extract (300 mg) of Rhizoma Coptidis: scoulerine (3.6 mg, mass fraction 71%) and isocorydine 9.2 mg, mass fraction 92%), dehydrocheilanthifoline (5.5 mg, 85%), dehydrocavidine (7.5 mg, 76%), palmatine (20.4 mg, , Mass fraction: 97%). Conclusion: The preparation of alkaloids from Coptis chinensis Franch by high-speed countercurrent chromatography is time-saving, convenient and high yield and purity.
