目的:观察SD大鼠角朊细胞在体外培养的生长特性。方法:采用dispase酶消化法从SD大鼠仔鼠皮肤获得角朊细胞,在体外进行培养并传代,采用倒置显微镜和MTT法观察细胞的形态学变化和生长曲线。结果:通过酶消化法可以获得较纯的角朊细胞,原代培养10d细胞即可以达到融合,传代后细胞部分发生变性,呈成纤维细胞样,传代次数高,成纤维样细胞比例也相应增高。结论:SD大鼠角朊细胞可以在体外培养分裂增殖,但传代后即发生变
Objective: To observe the growth characteristics of keratinocytes cultured in vitro in SD rats. Methods: Keratinocytes were obtained from the skin of pups of SD rats by dispase digestion method. The keratinocytes were cultured and passaged in vitro. The morphological changes and growth curves were observed by inverted microscope and MTT. Results: Pure keratinocytes could be obtained by enzymatic digestion. The cells were primary cultured for 10 days to reach confluency. After subculture, the degenerated cells were fibroblast-like with high passage times and higher proportion of fibroblast-like cells . Conclusion: The keratinocytes of SD rats can be cultured and proliferated in vitro, but they change after passage
