目的:观察荷瘤鼠肿瘤放疗后肿瘤组织内微血管密度(microvesseldensity,MVD)及肿瘤细胞血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)表达阳性率的变化,分析放疗对肿瘤血管生成的抑制作用。方法:24只荷B16黑色素瘤的C57BL/6小鼠随机分为对照组和放疗组(30Gy/10次/10天),放疗结束后第5天处死小鼠,免疫组化法测定肿瘤组织内MVD值和肿瘤细胞VEGF表达阳性率,比较两组间的差异。结果:两组肿瘤组织内MVD值分别为52.7±9.1和26.4±5.5(t=8.52,P<0.001),两组肿瘤细胞VEGF表达阳性率分别为31.4%±6.5%和10.6%±3.0%(对照组VS放疗组)(t=10.1,P<0.001),均有明显显著性差异。结论:放疗可明显抑制荷瘤鼠肿瘤的血管生成过程。
Objective: To observe the changes of microvessel density (MVD) and the positive rate of vascular endothelial growth factor (VEGF) expression in tumor-bearing mice after radiotherapy, and analyze the inhibitory effect of radiotherapy on tumor angiogenesis. Methods: Twenty-four C57BL/6 mice bearing B16 melanoma were randomly divided into control group and radiotherapy group (30 Gy/10 times/10 days). The mice were sacrificed on the 5th day after the end of radiotherapy. Immunohistochemical method was used to determine the tumor tissues. The MVD value and the positive rate of VEGF expression in tumor cells were compared between the two groups. Results: The MVD values in the two groups of tumor tissues were 52.7±9.1 and 26.4±5.5, respectively (t=8.52, P<0.001). The positive rates of VEGF expression in tumor cells of the two groups were 31.4%±6.5% and 10.6%±3.0%, respectively ( The control group VS radiotherapy group (t=10.1, P<0.001), there were significant differences. Conclusion: Radiotherapy can significantly inhibit the angiogenesis of tumor-bearing mice.
