日的:探讨非霍奇金淋巴瘤(non Hodgkin lymphoma,NHL)(CR_1密度基因多态性及与淋巴细胞和红细胞免疫功能的相关性。方法:96例初治的NHL患者及60例正常献血者,采用PCR加HindⅢ酶切方法测定CR_1密度基因多态性分布;采用ELISA法测定患者血清中CD4和CD8分子的含量;采用郭峰法测定患者血液中RBC-C3bRR、RBC-ICR和TRR水平。结果:(1)NHL患者红细胞CR_1密度基因组HH型比率明显低于正常人群,而HL型及LL型比率都明显高于正常人群(P<0.01);(2)NHL患者红细胞CR_1密度基因HH型组RBC-C3bRR、TRR、CD4、CD4/CD8比值水平明显高于HL型组(P<0.05,P<0.01,P<0.05,P<0.01),而低于正常对照组HH型组分别为(P<0.01,P<0.01,P<0.05,P<0.01),并且LL型组低于HL型组分别(P<0.05,P<0.01,P<0.01,P<0.01);(3)NHL患者HH型组RBC-ICR明显低于HL型组(P<0.05)而高于正常对照组HH型组(P<0.05),并且LL型组高于HL型组(P<0.01);(4)NHL患者红细胞CR_1密度基因分布与红细胞及淋巴细胞免疫功能成正相关(P<0.01)。结论:NHL患者红细胞CR_1密度基因组HH型比率明显低于正常人群,而HL型及LL型比率都明显高于正常人群(P<0.01);CR_1密度基因多态性与机体的红细胞及淋巴细胞免疫功能相关。
Day: To investigate the relationship between polymorphism of CR_1 density gene and lymphocyte and erythrocyte immune function in non-Hodgkin lymphoma (NHL) .Methods: 96 untreated NHL patients and 60 normal donors , PCR and Hind Ⅲ restriction enzyme digestion method was used to determine the density distribution of CR_1 gene. The contents of CD4 and CD8 in sera of patients were determined by ELISA. The levels of RBC-C3bRR, RBC-ICR and TRR in blood were measured by Guo Feng method. Results: (1) The ratio of HH type in erythrocyte CR1 density genotype of NHL patients was significantly lower than that of normal people, while the ratio of HL type and LL type were significantly higher than that of normal people (P <0.01). (2) The ratio of RBC-C3bRR, TRR, CD4 and CD4 / CD8 in HH group was significantly higher than that in HL group (P <0.05, P <0.01, P <0.05, P <0.01) (P <0.01, P <0.05, P <0.01) in the HH group and the LL group were lower than those in the HL group (P <0.01). 0.05, P <0.01, P <0.01, P <0.01); (3) The RBC-ICR of HH group was significantly lower than that of HL group (P <0.05) Group HH group (P <0.05), and LL group was higher than HL group (P <0.01). (4) The distribution of erythrocyte CR_1 density in NHL patients was positively correlated with the immune function of erythrocyte and lymphocyte (P0.01) .Conclusion: The ratio of HH genotypes in erythrocyte CR_1 Lower than the normal population, while the ratio of HL type and LL type were significantly higher than the normal population (P <0.01). The density polymorphism of CR_1 was related to the immune function of erythrocytes and lymphocytes.
