目的检测HepG2细胞脂肪变模型中caspase-3活性的变化,探讨抗氧化剂维生素E(VitE)对其的影响。方法 0.5 mmol/L和1 mmol/L的混合脂肪酸分别干预HepG2细胞,在不同时间点检测细胞内caspase-3活性。将1 mg/L的VitE干预脂肪变性HepG2细胞,在不同时间点检测caspase-3活性的变化。结果脂肪酸组HepG2细胞caspase-3活性较正常对照组显著升高(P<0.05),VitE干预组细胞caspase-3活性均较脂肪酸组明显下降(P<0.05)。结论脂肪酸可诱导HepG2细胞caspase-3活性升高,抗氧化剂VitE能下调脂肪变性HepG2细胞caspase-3活性从而减轻细胞损伤,这可能与氧化应激和内质网应激的改善相关。
Objective To detect the changes of caspase-3 activity in HepG2 cell steatosis model and investigate the effect of vitamin E on it. Methods HepG2 cells were treated with 0.5 mmol / L and 1 mmol / L mixed fatty acids respectively, and the activity of caspase-3 was detected at different time points. HepG2 cells were treated with VitE at a concentration of 1 mg / L, and the changes of caspase-3 activity were detected at different time points. Results The caspase-3 activity of HepG2 cells in fatty acid group was significantly higher than that in normal control group (P <0.05). The activity of caspase-3 in VitE group was significantly lower than that in fatty acid group (P <0.05). Conclusion Fatty acid can induce caspase-3 activity in HepG2 cells. Antioxidant VitE can down-regulate the caspase-3 activity in HepG2 cells, which may be related to oxidative stress and the improvement of endoplasmic reticulum stress.
