本研究从水芹中克隆得到一个HSP基因Oj HSP90,生物信息学分析表明:水芹Oj HSP90基因全长2 100 bp,编码699个氨基酸;Oj HSP90蛋白的理论分子量为80.3 k D,等电点为5.01,是一种亲水性的稳定蛋白;Oj HSP90蛋白的二级结构中,α-螺旋占52.93%,延伸链占16.74%,β-折叠占5.87%,无规则卷曲占24.46%;该蛋白的N端含有1个HSP90超家族蛋白保守的结构域YSNKEIFLRE,能够与ATP结合;进化树分析表明,Oj HSP90蛋白与拟南芥At HSP90蛋白和烟草Nt HSP90的相似度最高,分别为91.99%和92.13%。本研究旨在为进一步研究HSP90基因的功能以及研究水芹耐高温途径提供理论参考。
In this study, an HSP gene Oj HSP90 was cloned from cress. Bioinformatics analysis showed that the Oj HSP90 gene was 2 100 bp in length and encoded 699 amino acids. The theoretical molecular weight of Oj HSP90 protein was 80.3 kD and its isoelectric point Was 5.01, which was a hydrophilic stable protein. The secondary structure of Oj HSP90 protein was 52.93% in α-helix, 16.74% in extension chain, 5.87% in β-sheet and 24.46% in random coil. The N-terminus of the protein contains one conserved domain YSNKEIFLRE of HSP90 superfamily, which can bind to ATP. The phylogenetic tree analysis showed that the similarity between Oj HSP90 protein and Arabidopsis At HSP90 protein and tobacco Nt HSP90 were 91.99% And 92.13%. The purpose of this study is to provide a theoretical reference for further study of the function of HSP90 gene and the study of its pathogenicity.
