目的:检测广西玉林市药材市场上流通的不同规格等级天麻中天麻素和总天麻素的含量。方法:采用反相高效液相色谱法,色谱柱为Agilent C18液相色谱柱(150mm×4.6mm,5μm),检测波长为220nm,流速为1mL/min,柱温为35℃,流动相为乙腈-0.05%磷酸溶液(3∶97)。结果:天麻素在0.24~4.8μg进样量范围内与峰面积呈良好的线性关系,r=0.999 9,天麻素和总天麻素的平均回收率分别为100.43%、100.69%。24批市售天麻的天麻素含量为0.27%~1.47%,均超过2015版药典中0.25%的标准;总天麻素含量为1.97%~5.50%。结论:市售24批天麻样品中的天麻素含量均符合2015版药典要求;以总天麻素含量为评价指标对天麻进行品质评价的方法较药典以游离天麻素含量为指标更为合理。
Objective: To detect the content of gastrodin and total gastrodin in different grades of Tianma which circulated on the market of Yulin, Guangxi. Methods: Reversed-phase high performance liquid chromatography (HPLC) with Agilent C18 column (150mm × 4.6mm, 5μm), detection wavelength of 220nm, flow rate of 1mL / min, column temperature of 35 ℃ and mobile phase of acetonitrile -0.05% phosphoric acid solution (3:97). Results: Gastrodin showed a good linear relationship with the peak area in the range of 0.24-4.8 μg injection volume, r = 0.999 9. The average recoveries of gastrodin and total gastrodin were 100.43% and 100.69%, respectively. 24 batches of gastrodin gastrodin content of 0.27% to 1.47%, all exceed the 2015 version of the Pharmacopoeia 0.25% standard; total gastrodin content of 1.97% to 5.50%. Conclusion: The gastrodin content in 24 batches of gastrodia samples are all in line with the Pharmacopoeia of 2015. The method of evaluating the gastrodin quality with total gastrodin content is more reasonable than Pharmacopoeia.
