目的探讨三丁酸甘油酯(tributyrin,TB)对人急性髓系白血病HL-60细胞及胃癌细胞(SGC-7901)的增殖、细胞周期的调控及细胞凋亡的影响,并比较TB对HL-60细胞及SGC-7901的作用效果。方法用MTT法测定TB在不同浓度、不同时间的细胞增殖变化;应用免疫组化检测抑癌基因p16的表达;应用流式细胞仪观察细胞周期;通过DNA电泳观察细胞凋亡情况。结果TB可以抑制HL-60细胞及SGC-7901细胞增殖,其抑制率与药物浓度和作用时间呈依赖关系;1·0mmol/L的TB对HL-60细胞作用72h,有典型的DNA梯形条带。流式细胞仪分析证实TB使细胞周期阻滞在G0/G1期。结论TB对HL-60细胞的抑制作用更明显,其作用机制可能与上调Bmi-1基因上的p16INK4蛋白表达有关。
Objective To investigate the effects of tributyrin (TB) on the proliferation, cell cycle regulation and apoptosis of human acute myeloid leukemia HL-60 cells and gastric cancer cells (SGC-7901), and compare TB to HL- 60 cells and SGC-7901 effect. Methods The cell proliferation of TB was measured by MTT assay at different concentrations and at different times. The expression of tumor suppressor gene p16 was detected by immunohistochemistry. The cell cycle was observed by flow cytometry. The apoptosis was observed by DNA electrophoresis. Results TB could inhibit the proliferation of HL-60 cells and SGC-7901 cells. The inhibition rate was dependent on the drug concentration and time. The effect of TB on the HL-60 cells was 72 h, and there was a typical DNA trapezoidal band. . Flow cytometry analysis confirmed that TB arrested the cell cycle in G0/G1 phase. Conclusion The inhibitory effect of TB on HL-60 cells is more obvious. The mechanism may be related to up-regulation of p16INK4 protein expression on Bmi-1 gene.
