目的:以五味子为原料,以总木脂素为研究对象,展开了对总木脂素的提取、分离纯化及还原能力的研究。方法:本文通过静态和动态实验对5种大孔树脂进行筛选,以吸附、解析数据为指标,优选大孔树脂纯化工艺,并利用Fe3+法测定总木脂素体外还原能力。结果:确定AB-8型树脂最优,最佳工艺条件为:五味子总提取物上样浓度为1.5 mg/m L,流速为1 BV/h;最终用50%乙醇(低浓度洗脱剂)和70%乙醇(高浓度洗脱剂)进行洗脱;体外还原能力结果显示:五味子总木脂素在50%和70%乙醇组分对Fe3+表现出较高的还原能力。试验结果显示,通过大孔树脂纯化五味子总木脂素及其体外还原能力的研究,五味子总木脂素的含量从11.68%提高到41.48%。结论:本实验的梯度洗脱法可显著提高五味子中总木脂素的质量分数,并且五味子总木脂素在50%和70%乙醇组分表现出良好的体外还原能力,为今后五味子总木脂素研究提供理论依据。
OBJECTIVE: To study the extraction, separation, purification and reducing ability of total lignans from Schisandra sphenanthera as raw material and total lignans as the research object. Methods: Five kinds of macroporous resins were screened by static and dynamic experiments. The adsorption and desorption data were used as indexes. The macroporous resin purification process was optimized. The in vitro reduction of total lignans was determined by Fe3 + method. Results: The optimal AB-8 resin was determined. The optimum conditions were as follows: the total concentration of Schisandra chinensis fruit extract was 1.5 mg / m L and the flow rate was 1 BV / h; finally, 50% ethanol (low concentration eluent) And 70% ethanol (high concentration of eluent). The results of in vitro reduction showed that 50% and 70% ethanol components of Schisandra sp. Showed higher reducibility for Fe3 +. The results showed that the total lignan content of Schisandra chinensis increased from 11.68% to 41.48% by purifying total lignans from macroporous resin and its in vitro reduction ability. Conclusion: The gradient elution method can significantly improve the mass fraction of total lignans in Schisandra chinensis and the total lignans of Schisandra chinensis exhibits good in vitro reduction ability in 50% and 70% ethanol fractions, Lipid research to provide a theoretical basis.
