目的:应用大孔吸附树脂结合聚酰胺柱色谱法富集分离独一味中总黄酮、总环烯醚萜苷类极性成分,实现工业化生产。方法:用系列色谱设备进行分离富集,一阶导数紫外分光光度法测定总环烯醚萜苷含量、高效液相色谱法测定山栀苷甲酯,8-O-乙酰山栀苷甲酯,并考察不同洗脱条件下有效成分总环烯醚萜苷的变化。结果:采用70%乙醇溶液为溶媒。大孔吸附树脂柱用“下进上出”的进样方式和“上进下出”的洗脱方式时,得率为20.80%,总环烯醚萜苷、山栀苷甲酯和8-O-乙酰山栀苷甲酯的转移率分别为70.77%,76.36%,68.74%。结论:生产工艺方法及参数可以成功地将实验成果转化为工业化生产。
OBJECTIVE: To enrich and separate the polar components of total flavonoids and total iridoid glycosides from the monosodium glutamate by macroporous adsorption resin and polyamide column chromatography, and to achieve industrialized production. Methods: The total iridoid glycosides were determined by a series of chromatographic equipment, the first derivative UV spectrophotometry was used to determine the content of total iridoid glycosides. The contents of methyl and 8-O-methylglucoside methyl ester were determined by high performance liquid chromatography. The changes of the total iridoid glycosides of the active ingredients under different elution conditions were investigated. Results: 70% ethanol solution was used as vehicle. Macroporous resin column with “down on the ” injection mode and “up and down ” elution mode, the yield was 20.80%, the total iridoid glycosides, androsterone methyl ester And 8-O-acetylglucoside methyl ester transfer rates were 70.77%, 76.36%, 68.74%. Conclusion: The production process and parameters can successfully convert the experimental results into industrialized production.
