目的:探讨吲哚乙酸(IAA)联合辣根过氧化物酶(HRP)对BXPC3细胞凋亡及自由基表达的影响.方法:台盼蓝(曲利本兰)排斥试验,研究IAA/HRP对BXPC3细胞死亡率的影响;原位细胞凋亡检测法检测细胞凋亡;生化法检测细胞内SOD,MDA的含量;激光共聚焦显微镜检测细胞内的自由基含量.结果:与对照组相比,IAA/HRP组的细胞凋亡数增加,而且随着IAA浓度的增大,细胞凋亡数存在上升的趋势;SOD活性和MDA含量在IAA/HRP组中均明显升高;对照组的自由基含量比IAA/HRP组明显降低,且自由基的含量具有浓度和时间依赖性.结论:IAA联合HRP诱导自由基产生是诱发人胰腺癌BXPC3细胞凋亡的可能机制之一.
OBJECTIVE: To investigate the effects of IAA and HRP on the apoptosis and the expression of free radicals in BXPC3 cells.Methods: Trypan blue exclusion test was used to investigate the effects of IAA / HRP BXPC3 cells were detected by flow cytometry.Apoptosis was detected by in situ cell apoptosis assay.Cytochemical assay was used to detect the content of intracellular SOD and MDA.Results: Compared with the control group, The number of apoptotic cells in IAA / HRP group increased, and with the increase of IAA concentration, the number of apoptotic cells tended to increase; the activity of SOD and MDA content in IAA / HRP group were significantly increased; the free radicals The contents of free radicals were significantly lower than that of IAA / HRP group, and the content of free radicals was concentration- and time-dependent.Conclusion: IAA combined with HRP induces free radical production is one of the possible mechanisms of inducing apoptosis of human pancreatic cancer BXPC3 cells.
