采用甲醇提取油菜蜂花粉样品中黄酮类物质,利用高效液相色谱技术,优化了实验提取和分离测定条件。采用ODS C18色谱柱,乙腈-0.5%磷酸流动相,梯度洗脱,检测波长为260 nm。通过对油菜蜂花粉活性成分槲皮素的检测,确定该方法在0.02~0.16 mg/mL范围内线性关系良好,回收率在92%~96%之间。通过对不同地区10个油菜蜂花粉样品的实验研究,运用统计学方法,计算了10个样品的相对保留时间和相对标准偏差,初步确定了油菜蜂花粉的18个共有峰和重叠率,建立了油菜蜂花粉指纹图谱,为油菜蜂花粉的质量控制提供依据。
The flavonoids in rape bee pollen samples were extracted by methanol, and the conditions of experimental extraction and separation were optimized by HPLC. An ODS C18 column was used with an acetonitrile-0.5% phosphoric acid mobile phase and a gradient elution with a detection wavelength of 260 nm. Through the detection of quercetin, the active ingredient of rape bee pollen, it was determined that the method had good linearity in the range of 0.02-0.16 mg/mL, and the recovery rate was between 92% and 96%. Through experimental research on 10 rapeseed pollen samples in different regions, statistical methods were used to calculate the relative retention time and relative standard deviation of 10 samples. The 18 common peaks and overlapping rates of rape bee pollen were preliminarily determined. Rape bee pollen fingerprinting provides a basis for quality control of rape bee pollen.
