目的评估在不同温度、时间条件下,二甲亚砜和乙二醇对人GV、MⅠ、未受精MⅡ卵子的毒性。方法收集本中心废弃的GV、MⅠ及未受精MⅡ卵子,并随机分配到A、B及C组。A组:将卵子于不同温度下放入ES中10 min或20 min。B组:将卵子于不同温度下放入VS中1 min或5 min;C组为对照组。统计分析卵子的发育潜能。结果 A组或B1、B2组中各种卵子的发育潜能没有显著差异(P>0.05),而MⅠ卵子的体外成熟率都显著低于C组(P<0.05)。B3、B4两组中GV卵子的体外成熟率显著低于C组(P<0.05)。B3组中MⅠ、未受精MⅡ卵子的成活率要显著高于B4组(P<0.05)。B3、B4两组中MⅠ卵子的体外成熟率、受精率显著低于C组,MⅡ卵子的受精率、囊胚率显著低于C组(P<0.05)。结论室温或37℃条件下,亚砜和乙二醇混合剂对GV、MⅠ或未受精MⅡ卵子的毒性无明显差异。
Objective To evaluate the toxicity of dimethyl sulfoxide and ethylene glycol to human GV, M Ⅰ and unfertilized M Ⅱ eggs under different temperature and time. Methods The abandoned GV, MⅠ and unfertilized MⅡ oocytes were collected and randomly assigned to A, B and C groups. Group A: The eggs were placed in ES at different temperatures for 10 min or 20 min. Group B: The eggs were placed in VS at different temperatures for 1 min or 5 min; Group C was the control group. Statistics analysis of ovum developmental potential. Results There was no significant difference in the developmental potential of various eggs between group A and group B1 and group B2 (P> 0.05), but the maturation rate of M Ⅰ oocytes was significantly lower than that of group C (P <0.05). The in vitro maturation rate of GV oocytes in B3 and B4 groups was significantly lower than that in C group (P <0.05). The survival rates of MⅠ and unfertilized MⅡ oocytes in group B3 were significantly higher than those in group B4 (P <0.05). The in vitro maturation rate and fertilization rate of MⅠ oocytes in B3 and B4 groups were significantly lower than those in C group. The fertilization rate and blastocyst rate of M Ⅱ oocytes were significantly lower than those in C group (P <0.05). Conclusion The toxicity of sulfoxide and ethylene glycol mixture to GV, MⅠ or unfertilized MⅡ oocytes was not significantly different at room temperature or 37 ℃.
