目的 为研究南方红豆杉芽愈伤组织诱导条件、最适培养基组成成分和紫杉醇含量的积累。方法 以南方红豆杉芽为外植体 ,在 MS培养基上 ,试验不同种类、不同浓度的激素及其组合对其愈伤组织诱导的影响。结果 芽诱导的最适培养基是 MS添加 2 ,4 - D2 .0 mg/L 和 NAA0 .5 mg/L;对愈伤组织的增殖、驯化和紫杉醇含量的积累则以 B5添加 2 ,4 - D 0 .5 mg/L、NAA 1.0 m g/L和 KT 0 .5 mg/L为最佳。结论 本研究为植物组织细胞培养生产紫杉醇提供实验依据。
Objective To study the callus induction conditions, composition of the optimum medium and paclitaxel content accumulation of Taxus chinensis var. mairei buds. Methods The buds of Taxus chinensis were used as explants to test the effects of different kinds and concentrations of hormones and combinations on callus induction on MS medium. Results The most suitable medium for shoot induction was MS supplementation 2, 4 - D2 0 mg/L and NAA 0.5 mg/L; the proliferation, acclimation and paclitaxel content accumulation of callus were supplemented with B5 2 ,4- D 0 .5 mg/L, NAA 1.0 mg/L and KT 0.5 mg/L were the best. Conclusion This study provides experimental basis for the production of taxol in plant tissue culture.
