目的利用7,12-二甲基苯蒽(DMBA)诱导大鼠绝经后乳腺癌模型,探讨染料木黄酮(Genistein)联合阿那曲唑(Anastrozole)对乳腺癌组织的抑制作用及机制。方法建立DMBA诱导的绝经后乳腺癌大鼠模型,随机分为对照组、染料木黄酮组、阿那曲唑组、染料木黄酮与阿那曲唑联合治疗组(简称联合治疗组),观察各组肿瘤生长情况,测定肿瘤增殖指数、凋亡指数;ELISA法检测雌二醇(E2)及RTPCR法检测17β-羟甾脱氢酶1(17β-HSD1)mRNA的水平。结果染料木黄酮和阿那曲唑单独治疗组肿瘤生长均受到抑制,联合治疗组的抑瘤效果更显著(P<0.05)。与对照组比较,联合治疗组的E2和17β-HSD1mRNA水平下降更明显(P<0.05)。结论染料木黄酮可抑制绝经后大鼠中乳腺肿瘤的生长,与阿那曲唑联合应用显示出降低E2和17β-HSD1水平的协同作用,可有效抑制乳腺肿瘤生长。
OBJECTIVE: To investigate the inhibitory effect of genistein combined with Anastrozole on breast cancer and its mechanism by using 7,12-dimethylbenzyl anthracene (DMBA) to induce postmenopausal breast cancer in rats. Methods The rat model of postmenopausal breast cancer induced by DMBA was established and randomly divided into control group, genistein group, anastrozole group, genistein combined with anastrozole group (the combined treatment group) The proliferation and apoptotic index of tumor were detected by ELISA. The level of 17β-hydroxydopanhydride 1 (17β-HSD1) mRNA was detected by ELISA and estradiol (E2) and RTPCR respectively. Results Both genistein and Anastrozole alone inhibited tumor growth, and the combination therapy group had more significant antitumor effects (P <0.05). Compared with the control group, the combination therapy group E2 and 17β-HSD1 mRNA levels decreased more significantly (P <0.05). Conclusions Genistein can inhibit the growth of breast tumors in postmenopausal rats. The combination of anastrozole and anastrozole shows the synergistic effect of decreasing the levels of E2 and 17β-HSD1, which can effectively inhibit the growth of breast tumors.
