目的建立高效毛细管电泳法分离盐酸尼卡地平等6种药物对映体。方法采用未涂层熔融石英毛细管柱,30 mmol.L-1磷酸盐缓冲液作为背景电解质,手性选择剂为羧甲基-β-环糊精(CM-β-CD)。结果在最佳条件下,盐酸尼卡地平、盐酸曲马多、酮康唑、氧氟沙星、阿替洛尔、盐酸特布他林的分离度(R)分别为3.44、1.50、2.37、7.50、1.83和2.80。结论 CM-β-CD对6种药物对映体均有较好的对映体选择性。该分离方法操作简单,重现性好。
OBJECTIVE To establish a high performance capillary electrophoresis method for the separation of six enantiomers of nicardipine hydrochloride. Methods Uncoated fused silica capillary column, 30 mmol.L-1 phosphate buffer as background electrolyte and chiral selective agent as carboxymethyl-β-cyclodextrin (CM-β-CD) were used. Results Under the optimal conditions, the separation (R) of nicardipine hydrochloride, tramadol hydrochloride, ketoconazole, ofloxacin, atenolol and terbutaline hydrochloride were 3.44, 1.50, 2.37, 7.50, 1.83 and 2.80. Conclusion CM-β-CD has good enantioselectivity for all six enantiomers. The separation method is simple and reproducible.
