目的观察不同浓度柔红霉素(DNR)对Jurkat细胞生物学特性的影响。方法采用不同浓度的DNR作用Jurkat细胞,观察细胞的生长状况,在不同的时间点收获细胞,检测DNR作用前后Jurkat细胞生物学特性的改变。结果当0.5μmol/LDNR作用12h或18h,可以保证低死亡率的同时,细胞发生凋亡的比率最高;DNR抑制Jurkat细胞bcl-2、PCNA蛋白的表达,使更多细胞受阻于G0/G1期。结论在合适的DNR浓度及作用时间条件下,可能通过抑制Jurkat细胞bcl-2、PCNA蛋白的表达达到诱导凋亡、抑制增殖的作用。
Objective To observe the effects of different concentrations of daunorubicin (DNR) on the biological characteristics of Jurkat cells. Methods Jurkat cells were treated with different concentrations of DNR. The growth of Jurkat cells was observed. The cells were harvested at different time points to detect the changes of the biological characteristics of Jurkat cells. Results When 0.5μmol / L LDNR was applied for 12h or 18h, the rate of apoptosis was the highest while low mortality was observed. DNR inhibited the expression of bcl-2 and PCNA protein in Jurkat cells and blocked more cells in G0 / G1 phase . Conclusion Under appropriate DNR concentration and time, it is possible to induce apoptosis and inhibit proliferation by inhibiting the expression of bcl-2 and PCNA in Jurkat cells.
