目的 :为探讨酒精代谢产物乙醛对胚胎发育的作用 ,采用胚胎中脑细胞微团培养研究乙醛对其增殖和分化的影响。方法 :从 13天龄体节数 35 36的鼠胚胎中分离中脑细胞 ,施与不同剂量的乙醛于体外连续培养 5天后 ,测定乙醛对增殖和分化的影响 ,细胞的分化抑制表示有潜在致畸性。结果 :乙醛对中脑细胞增殖和分化的结果 ,显示 0 .1mM及以上剂量即能明显的抑制细胞的增殖分化过程 ,显示出乙醛能阻止神经形成和集落形成 ,其分化抑制剂量 (ID5 0 )和增殖抑制剂量 (IP5 0 )分别为 9.97mM和 6.69mM。结论 :根据的标准乙醛是一种非特异性的细胞增殖分化抑制剂 ,具有潜在的致畸性。
OBJECTIVE: To investigate the effect of alcohol metabolite acetaldehyde on embryonic development, the effect of acetaldehyde on the proliferation and differentiation of embryonic mesencephalic cells was investigated by using microtome culture of embryonic mesencephalic cells. METHODS: Midbrain cells were isolated from embryos of 35 36-day-old chickens at 35 days. Different concentrations of acetaldehyde were cultured in vitro for 5 days to determine the effect of acetaldehyde on proliferation and differentiation. The differentiation inhibition of cells showed Potential teratogenicity. Results: The results showed that acetaldehyde inhibited the proliferation and differentiation of midbrain cells, and showed that the inhibitory effect of acetaldehyde on the proliferation and differentiation of midbrain cells was significantly inhibited by acetaldehyde at the dose of 0.1 mM and above, indicating that acetaldehyde blocked neurogenesis and colony formation. 0) and proliferation inhibition dose (IP5 0) were 9.97 mM and 6.69 mM, respectively. Conclusion: The standard acetaldehyde is a non-specific inhibitor of cell proliferation and differentiation with potential teratogenicity.
