目的:探讨健脾补肺化痰方对幼龄哮喘大鼠肺组织中TGF-β_1、c-fos表达的影响。方法:将120只雄性SD幼龄大鼠,随机分为正常组4周(Z4)、8周(Z8)、12周(Z12),模型组4周(M4)、8周(M8)、12周(M12),健脾补肺化痰方组4周(J4)、8周(J8)、12周(J12),地塞米松组4周(D4)、8周(D8)、12周(D12),每组30只,采用卵白蛋白(OVA)雾化激发建立哮喘大鼠模型,用Masson染色法及Img-Pro6.0图像分析系统,分别测定上述时间段各组大鼠肺组织中WAt/Pbm、WAi/Pbm及WAm/Pbm等气道形态学参数,同时采用逆转录聚合酶链反应(RT-PCR)分别检测各组大鼠肺组织中TGF-β_1、c-fosm RNA的表达。结果:与正常组相比,模型组肺组织中TGF-β_1、c-fos的表达及WAt/Pbm、WAi/Pbm、Wam/Pbm等气道形态学参数均有不同程度升高(P<0.05);与模型组相比,各治疗组以上指标均有不同程度下调(P<0.05);健脾补肺化痰方组与地塞米松组比较,差异无显著性(P>0.05)。结论:健脾补肺化痰方可通过下调TGF-β_1、c-fos的表达实现对气道重塑的抑制。
Objective: To investigate the effects of Jianpi Bufei Huatan Fang on the expression of TGF-β 1 and c-fos in lung tissue of young asthmatic rats. Methods: A total of 120 male SD rats were randomly divided into four groups (Z4, Z8, Z12), M4 (M8), M8 M12, J4, J8, J12, D4, D8, 12 weeks (M12, D12), 30 rats in each group. Asthma rat model was induced by ovalbumin (OVA) atomization. Masson staining and Img-Pro6.0 image analysis system were used to determine the expression of WAt / Pbm, WAi / Pbm and WAm / Pbm, respectively. Meanwhile, the expression of TGF-β 1 and c-fosm RNA in the lung tissue of each group were detected by reverse transcription polymerase chain reaction (RT-PCR) Results: Compared with the normal group, the expression of TGF-β 1 and c-fos in the model group and the airway morphology parameters such as WAt / Pbm, WAi / Pbm and Wam / Pbm increased to some extent (P <0.05 ). Compared with the model group, the above indexes of the treatment groups were all decreased to different extents (P <0.05). There was no significant difference in the indexes of Jianpi Bufei Huatan Fang group and dexamethasone group (P> 0.05). Conclusion: Jianpi Bufei Huatan can inhibit airway remodeling by down-regulating the expression of TGF-β 1 and c-fos.
