目的观察声动力学疗法(SDT)对肿瘤细胞超微结构的影响,探讨超声激活血卟啉杀伤肿瘤细胞的机制。方法采用频率为2.0MHz,声强为1.5W cm2的超声,结合血卟啉(HpD)对离体培养的小鼠艾氏腹水瘤(EAT)细胞处理3min,利用透射电镜观察不同时间段取材的细胞超微结构的变化。结果超声激活血卟啉对EAT细胞超微结构的破坏程度随取材时间的延长而加剧,损伤位点主要集中在细胞膜、线粒体、内质网及细胞核上,同时还观察到一些细胞表现出明显的凋亡特征。结论超声激活血卟啉主要通过破坏细胞超微结构杀伤肿瘤细胞,部分通过诱导凋亡杀伤。
Objective To observe the effect of sonodynamic therapy (SDT) on the ultrastructure of tumor cells and to explore the mechanism of ultrasonically activating hematoporphyrin to kill tumor cells. Methods Ehrlichchian cystadenocarcinoma (EAT) cells cultured in vitro with hematoporphyrin (HpD) were treated with ultrasound at a frequency of 2.0 MHz and a sound intensity of 1.5 W cm2 for 3 min. Results The degree of destruction of ultrastructure of EAT cells by hematoporphyrin by ultrasound was aggravated with the extension of time. The sites of injury were mainly concentrated in the cell membrane, mitochondria, endoplasmic reticulum and nucleus. Some cells were also observed to be significantly Apoptosis characteristics. Conclusion Ultrasound-activated hematoporphyrin can kill tumor cells mainly by disrupting the ultrastructure of the cells and partly induce apoptosis.
