沙门氏菌/微粒体酶体外试验系统是一个灵敏、快速、经济、可靠的试验,用于检出环境中的诱变剂。它是利用组氨酸需要型鼠伤寒菌株,在诱变剂的作用下,使之发生回变为不需组氨酸型,从而能在最低营养培养基上长成菌落。由于鼠伤寒沙门氏菌缺乏哺乳类动物的药物代谢酶系统,故在必要时,试验中要加入哺乳动物(常用大鼠)的微粒体酶(常用肝)。在实际工作中,是应用肝匀浆9,000×g离心的上清液。其中除含微粒体膜结合酶外,还含其他的细胞内可溶性酶,如6-磷酸葡萄糖(G-6-P)脱氢酶。
Salmonella / microsomal in vitro assay system is a sensitive, rapid, economical and reliable assay for detection of mutagenic agents in the environment. It is the use of histidine-requiring murine typhoid strain, under the action of a mutagen to make it back to the non-histidine type, which can grow in the minimum nutrient medium colonies. Since S. typhimurium lacks a drug-metabolizing enzyme system in mammals, microsomal enzymes (commonly used in the liver) of mammals (commonly used rats) are added when necessary. In practice, the supernatant was centrifuged at 9,000 × g in liver homogenate. In addition to microsomal membrane-bound enzymes, it contains other intracellular soluble enzymes, such as glucose 6-phosphate dehydrogenase (G-6-P).
