目的 以肿瘤抗原P815AB抗原肽为模式抗原 ,研究CpG寡聚脱氧核苷酸 (CpGODN)对该肿瘤抗原表位肽的免疫佐剂效应。方法 用 [3H ] TdR掺入法研究CpGODN体外刺激小鼠脾细胞增殖效应 ,用ELISPOT和 [3H] TdR释放法检测不同构成形式的疫苗在体诱发特异性CTL应答的效应。结果 ODN182 6在体外可刺激小鼠脾细胞显著增殖。经 4次免疫 ,在“肽 +ODN182 6 +IFA”组中 ,4/6的小鼠体内出现了特异性CTL应答 ;在“肽 +ODN182 6 +PBS”组 ,有 3只小鼠在完成 4次免疫前先后死亡 ,余 3只中有 1只体内也可检测到相应CTL反应 ;而对照“肽 +IFA”组 ,6只小鼠体内均未诱发出特异性CTL应答。结论 CpGODN对P815AB表位肽有较为理想的免疫佐剂效应 ,但疫苗构成形式对其免疫效果有着决定性的作用 ,“肽 +ODN182 6 +IFA”是本研究中最有效的疫苗形式 ,同时需注意到CpGODN具有一定的毒副作用
Objective To study the immunoadjuvant effect of CpG oligodeoxynucleotide (CpGODN) on the tumor epitope peptide using the tumor antigen P815AB antigen peptide as a model antigen. Methods [3H] TdR incorporation method was used to study the effect of CpGODN on the proliferation of mouse spleen cells in vitro. The effects of different forms of vaccines on inducing specific CTL responses were detected by ELISPOT and [3H] TdR release assays. Results ODN182 6 could stimulate the proliferation of mouse spleen cells in vitro. After 4 immunizations, specific CTL responses occurred in 4/6 mice in the “Peptide + ODN182 6 + IFA” group; in the “Peptide + ODN182 6 + PBS” group, 3 mice completed 4 After the first immunization, the mice died one after another and one of the remaining three also had a corresponding CTL response. In the control “peptide + IFA” group, no specific CTL responses were induced in the six mice. Conclusion CpG ODN has an ideal immunoadjuvant effect on P815AB epitope peptide, but the vaccine composition has a decisive effect on its immune effect. “Peptide + ODN182 6 + IFA” is the most effective vaccine form in this study.
