目的:采用高效液相色谱(HPLC)建立了减肥类保健食品中盐酸西布曲明和酚酞的同时测定方法。方法:以乙腈提取样品中盐酸西布曲明和酚酞,测定时以Cloversil-C18(4.6 mm×150 mm,5μm)为分析柱,乙腈-己烷磺酸钠柠檬酸溶液(60∶40,v/v)为流动相,流速1.0 mL.min-1,检测波长为223 nm。结果:盐酸西布曲明和酚酞在0.001~0.1 mg.L-1范围内线性关系良好,相关系数(r)大于0.9993,盐酸西布曲明和酚酞最低检出浓度分别为5 mg.kg-1和2.5 mg.kg-1。减肥茶、减肥胶囊和减肥片的5.0,10,50 mg.kg-1的平均添加回收率均大于80%,RSD(n=6)小于10%。结论:本方法简便快速、灵敏度高、结果准确可靠,可实现2种成分同时测定,适用于减肥类保健品中盐酸西布曲明和酚酞的监督检验。
OBJECTIVE: To establish a method for the simultaneous determination of sibutramine hydrochloride and phenolphthalein in weight loss health food by high performance liquid chromatography (HPLC). Methods: Samples of sibutramine hydrochloride and phenolphthalein were extracted with acetonitrile. The samples were eluted with Cloversil-C18 (4.6 mm × 150 mm, 5 μm) and acetonitrile-hexanesulfonate citric acid solution (60:40, v / v) as the mobile phase at a flow rate of 1.0 mL.min-1 with a detection wavelength of 223 nm. Results: The linear correlation between sibutramine hydrochloride and phenolphthalein in the range of 0.001-0.1 mg.L-1 was good with a correlation coefficient (r) greater than 0.9993. The minimum detectable concentrations of sibutramine hydrochloride and phenolphthalein were 5 mg.kg-1 and 2.5 mg.kg-1. The average recoveries of dietary slimming tea, slimming capsule and slimming tablet at 5.0, 10 and 50 mg.kg-1 were both above 80% and RSD (n = 6) less than 10%. Conclusion: The method is simple and rapid with high sensitivity and accurate and reliable results. It can be used for the simultaneous determination of two components and is suitable for the supervision and inspection of sibutramine hydrochloride and phenolphthalein in weight loss health care products.
