目的观察天麻钩藤饮对自发性高血压大鼠(SHR)血清心脏基质金属蛋白酶-1(MMP-1)及基质金属蛋白酶抑制剂-1(TIMP-1)水平的影响。方法将WKY大鼠和SHR大鼠,随机分为天麻钩藤饮组、卡托普利组和SHR组,每组24只。观察大鼠的血压、左室重量指数、心肌组织胶原浓度、血管周围胶原面积与管腔面积比例及心肌胶原容积分数以及血清中心脏基质胶原酶-1及其抑制物的含量。结果天麻钩藤饮可使SHR大鼠的血压、Coll、LVI、PVCA、CVF明显降低,并能显著升高SHR组血清MMP-1水平(P<0.05),降低血清TIMP-1水平(P<0.05或P<0.01);卡托普利组则仅表现为降低血清TIMP-1水平(P<0.05),而对血清MMP-1水平则无明显作用(P>0.05)。结论天麻钩藤饮对SHR大鼠具有明显的降压及抑制心肌纤维化作用,能提升MMP-1水平并降低其抑制物TIMP-1的水平,从而促进心肌胶原的降解,可能是其逆转高血压心肌纤维化及左室肥厚的又一作用机制。
Objective To observe the effects of Tianma Gouteng Decoction on serum levels of matrix metalloproteinase-1 (MMP-1) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in spontaneously hypertensive rats (SHR) Methods WKY rats and SHR rats were randomly divided into Tianma Gouteng Decoction group, captopril group and SHR group, 24 rats in each group. The blood pressure, left ventricular mass index, myocardial collagen concentration, perivascular collagen area and lumen area ratio, myocardial collagen volume fraction, serum level of cardiac matrix collagenase-1 and its inhibitor were observed. Results Tianma Gouteng Yin could significantly decrease the blood pressure, Coll, LVI, PVCA and CVF in SHR rats and increase the level of serum MMP-1 (P <0.05) and serum TIMP-1 in SHR group (P < 0.05 or P <0.01). Captopril group only decreased serum TIMP-1 level (P <0.05), but had no effect on serum MMP-1 level (P> 0.05). Conclusion Tianma Gouteng Decoction has obvious antihypertensive effects and myocardial fibrosis in SHR rats, which can increase the level of MMP-1 and decrease the level of its inhibitor TIMP-1, thus promote the degradation of myocardial collagen, which may be reversed Myocardial fibrosis and left ventricular hypertrophy another mechanism of action.
