目的通过检测基质溶解因子(MMP-7)基因在直肠癌中的表达,探讨其在直肠癌进展中的作用。方法86对直肠癌肿瘤组织与正常直肠黏膜组织标本自身配对分成肿瘤组与正常组,提取总RNA,采用实时荧光定量逆转录-多聚酶链反应(Quantitative-Real-Time RT-PCR)法检测MMP-7基因的表达。结果MMP-7基因表达比值大于1的52例(62.7%,52/83),在肿瘤组的表达水平高于正常组(P<0.01)。MMP-7基因表达比值大于1的例数在Dukes C+D期与Dukes A+B期中分别为34例(72.3%,34/47)和18例(50.0%,18/36,P<0.05)。MMP-7基因表达与组织学分化程度(r=-0.635)和CEA水平(r=0.580)相关。结论MMP-7基因在直肠癌中的表达上调。MMP-7在直肠癌的浸润与转移中具有重要作用。
Objective To explore the role of matrix metalloproteinase-7 (MMP-7) in the progression of rectal cancer by detecting its expression in rectal cancer. Methods Tumor tissue samples and normal rectal mucosa samples from paracancerous tissues were paired and divided into tumor group and normal group respectively. Total RNA was extracted and quantified by Quantitative-Real-Time RT-PCR (Real-time RT-PCR) 7 gene expression. Results 52 cases (62.7%, 52/83) had MMP-7 gene expression ratio greater than 1, which was higher in the tumor group than in the normal group (P <0.01). 34 cases (72.3%, 34/47) and 18 cases (50.0%, 18/36, P <0.05) in the Dukes C + D stage and Dukes A + B stage, respectively. . The expression of MMP-7 was correlated with histological differentiation (r = -0.635) and CEA level (r = 0.580). Conclusion The expression of MMP-7 in rectal cancer is up-regulated. MMP-7 plays an important role in the invasion and metastasis of rectal cancer.
