目的:探讨羟基脲(hydroxyurea,HU)对大鼠生精细胞凋亡相关基因mRNA及蛋白表达水平的影响。方法:雄性大鼠,腹腔注射400 mg.kg-1HU,给药后6,12和24 h处死,取睾丸组织,分离总RNA和蛋白。结果:半定量反转录聚合酶链式反应(RT-PCR)和蛋白印迹(Western blot)分析凋亡相关基因、蛋白产物,显示HU处理后,Bax水平没有出现明显变化,但Bcl-2表达水平在HU处理后6或12 h明显降低;相反,FasL和Fas的表达水平在给药后12 h开始增加。结论:HU导致的大鼠睾丸生殖细胞的凋亡涉及Bcl-2蛋白和Fas信号系统。
Objective: To investigate the effect of hydroxyurea (HU) on mRNA and protein expression of apoptosis related genes in rat spermatogenic cells. Methods: Male rats were injected intraperitoneally with 400 mg.kg-1 UU and sacrificed at 6, 12 and 24 h after administration. The testis tissues were obtained and total RNA and protein were separated. Results: Apoptosis-related genes and protein products were analyzed by semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blotting. No significant changes in Bax levels were observed after treatment with HU, but Bcl- At 6 or 12 h after HU treatment, the level of FasL decreased obviously. On the contrary, the expression of FasL and Fas began to increase at 12 h after administration. Conclusion: HU induced apoptosis of testicular germ cells in rats involves Bcl-2 protein and Fas signaling system.
