RNA原位杂交技术是在组织显微水平研究目标基因时空表达分布的重要技术手段。在植物中,原位杂交技术广泛应用于拟南芥和水稻的研究,而在玉米的研究中该技术应用较晚,实验技术体系尚有待优化和完善。文章以玉米为材料探索和优化原位杂交实验技术体系,并着重对实验的方法步骤、技术要点、结果分析等方面进行讨论和总结。实验结果表明,优化后的实验技术体系能够特异的检测到目标基因在玉米雄花中表达分布。
RNA in situ hybridization is an important technique to study the spatio-temporal distribution of target genes at the microscopic level. In plants, in situ hybridization is widely used in Arabidopsis and rice research, but in the corn research late application of this technology, experimental technology system has yet to be optimized and perfected. The article explores and optimizes the in-situ hybridization experimental technology system using corn as the material, and emphatically discusses and summarizes the experimental method steps, technical points and result analysis. The experimental results show that the optimized experimental technology system can specifically detect the expression of the target gene in maize male flower.
