为了得到巨大芽孢杆菌(Bacillus megaterium)发酵的最佳培养基,在单因子试验的基础上,应用Plackett-Burman设计法对影响芽孢产量的基本培养基组分中的关键因子进行了筛选,并进一步采用响应面分析法(RSM)对影响芽孢产量的关键因素最佳水平作了深入的研究。结果表明,影响巨大芽孢杆菌芽孢产量的关键因素是豆粕和淀粉的浓度。通过RSM的拟合和推算得到在豆粕和淀粉浓度分别为48.85和19.67 g/L时,此时模型预测发酵最佳产量为43.16×108CFU/mL,验证值为42.2×108CFU/mL,预测值与验证值之间吻合较好,比原始培养基提高了约10倍。
In order to obtain the best medium for the fermentation of Bacillus megaterium, the key factors of the basic medium components affecting the spore yield were screened by the Plackett-Burman design method on the basis of single-factor experiments and further Response surface analysis (RSM) was used to investigate the optimal level of the key factors affecting spore yield.
