目的探讨甲状腺功能减退(甲减)对雄性大鼠睾丸线粒体性激素受体(ERα和AR)基因mRNA表达的影响,为研究甲减对雄性生殖系统损伤机制提供理论依据。方法 30只健康雄性wistar大鼠,随机分3组,按体质量,采用随机数字表法分为3组:对照组(C组,灌胃生理盐水1 ml/100 g)、低剂量组(L组,灌胃0.1 mg/kg丙硫氧嘧啶1 ml/100 g)和高剂量组(H组,灌胃10 mg/kg丙硫氧嘧啶1 ml/100 g),每组10只,灌胃60 d,每隔2天称体重1次。灌胃结束次日处死大鼠,放射免疫法检测血清中甲状腺激素(TT3、TT4和TSH);实时荧光定量PCR(RT-PCR)法检测睾丸线粒体ERα和AR基因mRNA相对表达量。结果与C组和L组比较,H组大鼠体重、TT3和TT4出现明显降低(F=19.82,P<0.05;F=139.34,P<0.05),TSH水平及睾丸脏器系数显著升高(F=60.389,P<0.05;F=21.83,P<0.05);基因结果显示:H组ERα基因mRNA相对表达量显著高于C组和L组(F=13.35,P<0.05),AR基因mRNA相对表达量显著性高于C组(F=4.561,P<0.05)上述差异均有统计学意义。结论甲减可影响性激素受体基因mRNA表达,进而损伤大鼠生殖系统。
Objective To investigate the effect of hypothyroidism (Hypothyroidism) on the mRNA expression of testicular mitochondrial ERα and AR in male rats, and to provide a theoretical basis for the study of hypothyroidism on the mechanism of male reproductive system injury.
