以提取的香鳞毛蕨总RNA为试材,采用RACE方法得到香鳞毛蕨psbA基因全长,研究其序列的生物信息学特征并提交序列至GeneBank。结果表明:克隆的基因全长1 425bp,开放阅读框为1 062bp,编码353个氨基酸,与其它植物相比具有很高的同源性,基因命名为DfpsbA,GeneBank登录号为KJ728648。该研究结果可为进一步阐明香鳞毛蕨光合作用机理、基因表达与调控奠定基础,同时也为植物分类与进化提供依据。
The total RNA extracted from A. fragrans was used as experimental material. The full-length psbA gene was amplified by RACE, the bioinformatics characteristics of its sequence were studied and the sequence was submitted to GeneBank. The results showed that the cloned gene was 1 425 bp in length and contained 1 062 bp in open reading frame (ORF) encoding 353 amino acids, which was highly homologous to other plants. The gene was named DfpsbA and the accession number of GeneBank was KJ728648. The results of this study may provide a basis for further elucidating the mechanism of photosynthesis, gene expression and regulation, and also provide basis for plant taxonomy and evolution.
