目的探讨miRNA-7(miR-7)对卵巢癌SKOV3细胞增殖及侵袭能力的影响。方法将卵巢癌SKOV3细胞分为miR-7抑制剂组、miR-7模拟物组和对照组,分别将miR-7抑制剂或miR-7模拟物转染至miR-7抑制剂组或miR-7模拟物组细胞,对照组细胞转染无关序列,Real-time PCR法检测各组细胞miR-7表达水平,分别用MTT方法与Transwell细胞侵袭实验检测转染后各组SKOV3细胞的增殖与侵袭能力变化。Real-time PCR与Western blot实验分别检测各组细胞EGFR mRNA和蛋白表达水平。结果与对照组相比,miR-7抑制剂组SKOV3细胞miR-7表达水平显著降低,miR-7模拟物组则显著升高(P<0.01)。与对照组相比,miR-7抑制剂组SKOV3细胞增殖与侵袭能力显著升高,EGFR mRNA及蛋白表达水平显著升高,miR-7模拟物组则均显著降低(P<0.01)。结论 miR-7抑制卵巢癌SKOV3细胞的增殖和侵袭可能与下调EGFR的表达相关。
Objective To investigate the effect of miRNA-7 on the proliferation and invasion of ovarian cancer SKOV3 cells. Methods SKOV3 cells were divided into miR-7 inhibitor group, miR-7 mimics group and control group. The miR-7 inhibitor or miR-7 mimics were transfected into miR-7 inhibitor group or miR- 7 mimics and control cells were transfected with unrelated sequences. Real-time PCR was used to detect the expression of miR-7 in each group. The proliferation and invasion of SKOV3 cells were detected by MTT assay and Transwell invasion assay Ability changes. Real-time PCR and Western blot were used to detect the expression of EGFR mRNA and protein in each group. Results Compared with the control group, miR-7 expression was significantly decreased in miR-7 inhibitor SKOV3 cells and significantly increased in miR-7 mimics (P <0.01). Compared with the control group, the proliferation and invasion ability of SKOV3 cells in miR-7 inhibitor group were significantly increased, the expression of EGFR mRNA and protein were significantly increased, while the miR-7 mimics group was significantly decreased (P <0.01). Conclusion miR-7 inhibits the proliferation and invasion of ovarian cancer SKOV3 cells may be related to the down-regulation of EGFR expression.
