激光共聚焦显微镜 (LSCM)是九十年代初出现的一种可用荧光标记进行微形态观察的高分辨显微镜 ,与传统方法比较 ,具有精确、清晰、动态和三维成像等突出优点。骨计量学是研究代谢性骨病的重要方法。本研究将激光共聚焦显微技术引入骨计量学中 ,达到了精确定位和定量分析骨微结构的目的 ,具有无须染色 ,对厚片可进行荧光染色观察的特点 ,其清晰度高 ,制片易 ,可与组织化学、免疫组织化学等方法结合 ,从多个角度、多个层次同时观察骨微结构与各种骨细胞的形态与功能。为代谢性骨病的研究和诊断提供了新的方法和途径。 Confocal laser scanning microscopy (LSCM) is a high-resolution microscopy microscope that can be used for micromorphological observation in the early 1990s. Compared with traditional methods, it has outstanding advantages such as precise, clear, dynamic and three-dimensional imaging. Bone metrology is an important method to study metabolic bone disease. In this study, laser confocal microscopy was introduced into bone metrology to achieve precise positioning and quantitative analysis of bone micro-structures. It has the characteristics of no need for staining, and can be observed with fluorescent staining of thick slices. Its high definition and production Easy, combined with histochemistry, immunohistochemistry and other methods, simultaneously observe the microstructure and function of bone micro-structure and various bone cells from multiple angles and at multiple levels. It provides new methods and approaches for the research and diagnosis of metabolic bone diseases.
