目的通过模拟计算及循证验证,探讨溶媒pH值对氨茶碱、氨溴索、拉贝洛尔、利托君4种药物理化性质及临床应用的影响,以期为临床实际工作提供理论指导及定量参考指标。方法通过Pubmed Compound查阅药物相关化学信息,查阅《马丁代尔药物大典》药物临床信息,应用ACD/LAB 6.0软件计算药物logP、pKa、logD及溶解度。并查阅药品说明书及临床研究进行循证验证。结果溶媒pH值对于4种药物的logD及溶解度均具有一定的影响,其变化趋势呈指数型变化。以上4种药物均可与葡萄糖注射液或0.9%氯化钠注射液配伍使用,但使用时的注意事项各有特点。结论溶媒pH值对药物溶解平衡具有一定的影响,药物化学性质的模拟计算可以为临床工作提供理论指导及定量参考数据,具有一定的借鉴意义。
OBJECTIVE: To study the effect of pH value on the physicochemical properties and clinical application of aminophylline, ambroxol, labetalol and ritodrine by simulation and evidence-based validation, in order to provide theoretical guidance for clinical practice and Quantitative reference index. Methods The drug related chemical information was obtained by Pubmed Compound. The clinical information of the drug was also reviewed. The logP, pKa, logD and solubility of the drug were calculated by ACD / LAB 6.0 software. And access to drug documentation and clinical research evidence-based verification. Results The pH value of the medium had some effects on the logD and the solubility of the four drugs, and the change tendency showed an exponential change. The above four kinds of drugs can be used with glucose injection or 0.9% sodium chloride injection compatibility, but when using the precautions have their own characteristics. Conclusion The pH value of the vehicle has a certain influence on the dissolution equilibrium of the drug. The simulation calculation of the chemical properties of the drug can provide theoretical guidance and quantitative reference data for clinical work, which is of certain reference significance.
