在过去华西1号(HX-1)液有效保存肝脏24小时,保存胰腺48小时的基础上,以高渗枸椽酸盐(HC-WCU)液为对照,采用狗自体肾移植模型观察其对狗肾脏保存效果。实验采用本地杂种狗20只,体重8~15kg,雌雄不限,随机分成①对照组(HC-WCU);②实验组(HX-1液组),各组又根据保存时间不同分为48,72小时两个亚组,每亚组动物各5只。切取狗左侧肾脏灌注后低温保存48,72小时行自体异位肾移植。同时切取右侧肾,观察移植术后狗血肌酐,存活率,形态学检查。实验发现1,HC-WCU液48小时组术后血肌酐峰值为642.60±158.60μmol/L,72小时组术后血肌酐峰值为686.20±154.04μmol/L。HX-1液48小时组术后血肌酐峰值为448.32±36.90μmol/L,72小时组术后血肌酐峰值为524±109.38μmol/L。2.HC-WCU肾保存液保存48,72小时移植存活率为80%和40%。HX-1液保存48,72小时移植存活率为100%和60%。3存活12天两组形态学无明显差异,HC-WCU液组死于肾衰的肾组织损害明显重于HX-1液组,尤以线粒体为甚。结论:HX-1液对狗肾脏的保存效果优于HC-WCU液,其有效保存时间达72小时,保存机制可能是防止细胞水肿,减轻细胞酸中毒,减少能量消耗,抗氧自由基,保护血管内皮细胞等。它作为腹部多器官保?
