目的 :探讨血管紧张素 (Ang )、内皮素 (ET- 1)对SHR、WKY VSMC增殖的作用。方法 :贴块法培养 SHR、WKY VSMC,用 3H- Td R参入和细胞计数反映 VSMC增殖情况。结果 :SHR、WKY VSMC3H- Td R参入随 Ang 、ET- 1浓度的增加而增加 ,呈剂量依赖性。 10 - 7mol/ L Ang 、ET- 1使SHR VSMC3H- Td R参入提高到对照组的 1.9、2 .7倍 ,WKYVSMC3H- Td R参入提高到对照组的 1.3、1.6倍。 Ang 不促使 SHR、WKY VSMC增生。 10 - 7mol/ L ET- 1分别使 SHR、WKY VSMC细胞数增加 2 0 9± 2 7%、97± 13%。 10 - 7mol/ LAng 和 ET- 1则使 SHR、WKY VSMC3H- Td R参入提高到对照组的 7.0、4.2倍 ,细胞数增加 3.0、1.3倍。结论 :Ang 、ET- 1对 VSMC的增殖有协同效应。高血压时 ,VSMC对促生长因子敏感性增加 ,更易发生肥大增殖
Objective: To investigate the effects of angiotensin (Ang) and endothelin (ET-1) on the proliferation of SHR and WKY VSMC. Methods: SHR and WKY VSMCs were cultured by patch method. The incorporation of 3H-TdR and cell counts were used to observe the proliferation of VSMCs. Results: The parameters of SHR and WKY VSMC3H-TdR increased with the increase of Ang, ET-1 concentration in a dose-dependent manner. The level of WKYVSMC3H-TdR increased to 1.3 and 1.6 times that of the control group by 10 -7 mol / L Ang and ET-1, respectively. Ang did not promote SHR, WKY VSMC hyperplasia. The numbers of cells in SHR and WKY VSMC increased by 2 0 9 ± 2 7% and 97 ± 13%, respectively, with 10 - 7 mol / L ET - 1. 10 - 7mol / LAng and ET - 1 increased the SHR and WKY VSMC3H - TdR concentrations to 7.0 and 4.2 times of the control group, respectively. The number of cells increased by 3.0 and 1.3 times. Conclusion: Ang, ET-1 has a synergistic effect on VSMC proliferation. Hypertension, VSMC increased sensitivity to growth factors, more prone to hypertrophy
