目的:根据已发表文献,综合评估尿苷二磷酸葡糖醛基转移酶1A7(UGT1A7)基因多态性与肿瘤易感性之间的关系。方法:按照统一的检索策略在相关中英文数据库中全面检索相关文献,对文献进行筛选、评价后获得相关研究的结果数据,然后在Stata10软件中用Meta分析的方法,计算合并的OR值及95%CI,并进行敏感性分析和发表偏倚的估计。结果:共有国内外17篇合格文献纳入本研究,累计病例和对照数分别为3081和5426例。合并结果显示,携带UGTA7*3等位基因者患肿瘤的危险性是携带野生型UGT1A7*1等位基因型的1.38倍(95%CI=1.18~1.62,P<0.001),携带UGTA7*2等位基因可能与肿瘤的危险性无关。亚组分析显示,UGT1A7*3等位基因型与肝癌和结直肠癌的危险性升高有关。发表偏倚评估未发现明显的偏倚,敏感性分析显示结论稳定可靠。结论:UGT1A7*3等位基因型可能与肿瘤的易感性升高有关。
OBJECTIVE: To evaluate the relationship between UGT1A7 gene polymorphism and tumor susceptibility according to published literature. Methods: According to a unified search strategy, we searched the relevant Chinese and English databases comprehensively, screened the literature, and obtained the data of the related research after evaluation. Then we used the Meta analysis method to calculate the OR and 95 % CI, and conduct sensitivity analysis and publish estimates of bias. Results: A total of 17 domestic and foreign literatures were included in this study. The total number of cases and controls were 3081 and 5426 respectively. The pooled results showed that the risk of tumor bearing the UGTA7 * 3 allele was 1.38-fold (95% CI = 1.18-1.62, P <0.001) with the wild-type UGT1A7 * 1 allele, carrying UGTA7 * 2 Bits may not be related to the risk of cancer. Subgroup analysis showed that the UGT1A7 * 3 allele was associated with an increased risk of liver cancer and colorectal cancer. No significant bias was found in publication bias assessments, and sensitivity analyzes showed a stable and reliable conclusion. Conclusion: The UGT1A7 * 3 allele may be related to the increased susceptibility to cancer.
