以甜叶菊无菌苗的叶片、茎段为外植体,对甜叶菊离体培养及再生体系的建立进行了试验研究。结果表明:以茎段为外植体进行甜叶菊的组织培养优于叶片外植体。茎段外植体愈伤组织诱导及丛生芽增殖的最适培养基为:MS+0.5mg/L6-BA+0.5mg/LNAA+3%蔗糖+0.7%琼脂;正交试验结果表明大量元素浓度对丛生芽的增殖影响最大,其次是6-BA、蔗糖,NAA的影响较小;生根最适培养基为MS+0.25mg/LIBA+0.2mg/LNAA+3%蔗糖+0.7%琼脂+0.3mg/L活性炭,添加低浓度活性炭的可明显提高生根率。
The leaves and stem segments of aseptic seedlings of Stevia rebaudiana were used as explants to study the in vitro culture of Stevia rebaudiana and the establishment of regeneration system. The results showed that the tissue culture of stevia was better than that of leaf explants using stem segments as explants.
